Renfrow attended Socastee High School in Myrtle Beach, South Carolina, where his father, Tim Renfrow, was the football coach. Renfrow was the quarterback for the football squad at Socastee. classified him as one of the two-star prospect personalities. Later, he had invitations to play for Appalachian State, Gardner-Webb, Presbyterian, and Wofford, all of which his father had attended during his football career. In 2015, Hunter Renfrow appeared in 13 games as a redshirt freshman. He appeared in ten games and accumulated 492 yards and five touchdowns. He threw two touchdown passes and was named the 2016 College Football Playoff National Champion.

Renfrow played 9 games as a sophomore in 2016, with some of those games being forfeited due to his injuries. He totaled 353 yards and four touchdown receptions. Renfrow caught a game-winning touchdown from quarterback Deshaun Watson with one second remaining in the 2017 College Football Playoff National Championship game against the Alabama Crimson Tide. Renfrow earned the Burlsworth Trophy in 2018, which is awarded to the greatest college players. He started off as a walk-on. Renfrow finished his undergraduate career with 49 receptions for 544 yards and one receiving touchdown, helping Clemson win its second national championship in three years.

Renfrow was taken by the Oakland Raiders in the fifth round of the 2019 NFL Draft as the 149th overall pick. He was picked 17th out of the available 28 wide raiders, one of three Clemson players taken by the Raiders, and the only offensive player drafted by the national championship team during the NFL draught. The NFL penalized Renfrow $15,000 on October 5, 2020. This was in response to the NFL fining or punishing him for attending a maskless charity event held by teammate Darren Waller during the COVID-19 outbreak.

Renfrow appeared in 16 games in 2020, beginning with the first six, and caught 56 receptions for 656 yards. He led the Dallas Cowboys to a 36-33 victory in Week 12 by passing for 8 receptions for 134 yards. Renfrow then raced for 9 receptions for 102 yards in a 17-15 victory over the Washington Football Team in Week 13. Renfrow rushed for 13 receptions for 117 yards and a 4-yard touchdown pass in a 9-48 defeat to the Kansas City Chiefs in Week 14. He had a career-high 103 receptions, 1,038 receiving yards, and 9 receiving touchdowns in 2021 as a regular season.
